There are 94 discoverable locations in Red Dead Redemption and all must be uncovered for 100% completion. Purchase maps from various stores to unlock them easily.
There are 9 counties in Red Dead Redemption which are displayed on the map from the start of the game. Once you visit a location inside them, they're usually checked off. There are 85 actual locations to visit in addition to the 9 counties and they'll be marked on your map when you get within a short distance of them.
There are 9 maps which can be purchased from General Stores (or the Gunsmiths in Escalera) and using the map in your Consumables menu will automatically mark the locations in that county onto your map.
The main benefit to discovering locations is the ability to set up a camp and fast travel to them. You're not able to fast travel to a waypoint in areas you haven't yet discovered.
The 94 locations are worth 6% towards 100% completion so you're awarded precicely 0.063829787234043% (rounded in-game) for each one you discover.
Discovering all locations will unlock the Trophy / Achievement "On the Trail of De Vaca".

Please Note: If you manually discover all locations in a county, the game will prevent you from purchasing the map. If a map is greyed out and "Not Available" in a store, it's likely you've already discovered everywhere in that county, so you can save your money.
You'll probably discover everywhere in Great Plains just by playing through the story and side missions so don't panic if the map can't be purchased.
